Combinatorial approaches: A new tool to search for highly structured β-hairpin peptides

Here we present a combinatorial approach to evolve a stable β-hairpin fold in a linear peptide. Starting with a de novo -designed linear peptide that shows a β-hairpin structure population of around 30%, we selected four positions to build up a combinatorial library of 20 4 sequences. Deconvolution of the library using circular dichroism reduced such a sequence complexity to 36 defined sequences. Circular dichroism and NMR of these peptides resulted in the identification of two linear 14-aa-long peptides that in plain buffered solutions showed a percentage of β-hairpin structure higher than 70%. The reaction between alkynes (I) and norbornadiene (II) affords the β-substituted Pauson—Khand adducts (III) as single regioisomers and the trifluoromethyl steering group can be easily removed in the presence of DBU and water.

The synthesis and characterization of twenty new pentamethylcyclopentadienyl-rhodium and iridium complexes containing N^N and N^O-chelating chloroquine analogue ligands are described. The in vitro antimalarial activity of the new ligands as well as the complexes was evaluated against the chloroquine sensitive (CQS) NF54 and the chloroquine resistant (CQR) Dd2 strains of Plasmodium falciparum. The antimalarial activity was found to be good to moderate; although all complexes are less active than artesunate, some of the ligands and complexes showed better activity than chloroquine (CQ). Phosphatidylcholine covalently linked to a methacrylate-based monolith as a biomimetic stationary phase for capillary liquid chromatography

Abstract In this study a strategy to immobilize phospholipids onto a polymer-based stationary phase is described. Methacrylate-based monoliths in capillary format (150 × 0.1 mm) were modified by soybean phosphatidylcholine through 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide coupling to obtain stationary phases suitable to mimic cell surface membranes. Fourier transform infrared (IR) spectroscopy in combination with multivariate data analysis is a versatile tool that can be applied to disease diagnosis. However, a rigorous validation of the obtained models is necessary in order to obtain robust results. Surfactant-Dependent Exciton Mobility in Single-Walled Carbon Nanotubes Studied by Single-Molecule Reactions

Measurements of stepwise photoluminescence quenching in individual, (n,m)-selected single-walled carbon nanotubes (SWCNTs) undergoing chemical reaction have been analyzed to deduce mobilities of optically generated excitons. For (7,5) nanotubes, the mean exciton range varies between approximately 140 and 240 nm for different surfactant coatings and correlates weakly with nanotube PL intensity. The results are consistent with a model of localized SWCNT excitons having substantial diffusional mobility along the nanotube axis.
